Morphometrical, cytological, and biochemical characterization of tungro-transmitter and tungro-nontransmitter populations of the green leafhopper, Nephotettix virescens (Distant)

Abstract
This study analysed and compared morphometrical, cytological, and biochemical variations in N. virescens tungro-transmitter and tungro-nontransmitter populations.
Discriminant analyses of morphometrical and cytological characteristics were employed to construct models that had greater than 50% probability of correctly classifying tungro-transmitters and tungro-nontransmitters.
Morphometrical investigations of ethanolfixed and KOH-cleared male adult N. virescens showed that tungro-transmitters had significantly greater values over tungronontransmiters for head width, long stylet length, penultimate rostral segment width, ultimate rostral segment width, ultimate rostral segment length, and vertex length and width.
Lacto-aceto-orcein squashes of testicular tissue of N. virescens revealed that tungro-transmitters had significantly higher values for leptonema and telophase I nuclei width, transmitters had greater relative chromosomes length for chromosomes I, V, IV, VIII, however tungro-nontransmitters had higher relative chromosome lengths for chromosomes II, III, VI, VII.
Electrophoretic analysis of genetic variation at seven enzyme loci showed that six were polymorphic and one was monomorphic. Hardy-Weinberg equilibria were obtained in three loci in tungro-transmitters, but only two in tungro-nontransmitters. Proportion of polymorphic loci, average number of alleles, and heterozygosity were similar in tungro-transmitters and tungro-nontransmitters. High genetic identity and low genetic distance indicated that both types of N. virescens belong to a single species.
