Abstract

The subchronic toxicity effects of Derris elliptica crude water extract on mature male albino rats were determined. Five treatment groups of five rats each were used and given the following doses of Derris: (1) High - 48 mg/kg; (2) Moderate - 32 mg/kg, (3) 16 mg/kg; and (4) Control - 0 mg/kg The last group contained rats treated with 0.047 mg/kg Racumin. Treatments were administered everyday for a period of 35 days using gavage method. Body weights increased gradually in the control and low-dose groups while decreased in the moderate and high doses groups from the third week onwards. Only the spleen and kidney weights were found to be significantly different across treatments. Histopathological evaluation of Derris-treated rats showed abnormalities in liver characterized by edema, and swollen and necrotic hepatocytes. Kidneys from the high dose treatment had atrophic glomeruli, while there was a dramatic spermatogenic arrest in the testes as well as interstitial fluid accumulation. No effects were observed in the spleen. A strong to very strong linear relationship was found to occur between the Denis doses and sperm counts, percentage of sperm abnormality, and mitotic cell incidence in the bone marrow. In the lone Racumin-treated rat which did not die in the course of the treatment period, congestion of sinusoids and degeneration of hepatocytes in the liver, glomerular atrophy and interstitial proliferation of eosinophilic cells in the kidney, and abnormal spermatogenesis were noted. However, results were not used for comparison with Derris-treated rats due to lack of replication.
