Abstract

Isozyme band pattern analysis of regenerants derived from the unirradiated and gamma irradiated embryogenie cultures of avocado, Persea americana Mill. was done using starch gel electrophoresis. Four enzyme systems used were acid phosphatase (ACPH), alkaline phosphatase (ALPH), esterase (EST) and 6-phosphogluconate dehydrogenase (PGD). ACPH and ALPH in avocado showed four zones of activity. EST and PGD, on the other hand, were observed in three and two zones, respectively. Among parental plants, variability was observed as reflected by the two types of band patterns exhibited by the three parental trees. Variability of band patterns was observed in the unirradiated and irradiated cultures. In ACPH, three band patterns were observed in the unirradiated regenerants and fourteen band patterns were observed in the irradiated regenerant. In ALPH, the unirradiated regenerants exhibited seven band patterns while the irradiated exhibited nine band patterns. In esterase, the unirradiated and irradiated showed nine band patterns. Three band patterns were exhibited by the two sample groups in PGD. Higher variability of band patterns was observed in the irradiated cultures since no two regenerants exhibited the same summarized band patterns. In the unirradiated regenerants, two pairs of regenerants and a group of three have the same band patterns. The higher variability of the irradiated regenerants may be attributed to the mutations induced by gamma irradiation and somaclonal variation. Percent polymorphic band in the four enzyme systems studied was 80% within the parent trees, 90% within the unirradiated tissue culture regenerants and 100% within the irradiated tissue culture regenerants. Considering all the sample groups studied, the 22 bands observed in the four enzyme systems are 100% polymorphic.
