Abstract

Meiosis in Dendrobium secundum f. album Valmayor and Tiu, Pomatocalpa bicolor (Lindl.) J. J. Smith, and Vanda lamellata var. boxalli Rchb.f. was studied using the acetocarmine squash technique. Flower buds were collected between 10am-12noon and fixed in Farmer's solution. Chromosome number, meiotic chromosome behavior, and pollen fertility were determined. In all cells at diakinesis in the three species, 19 Its were observed indicating a diploid chromosome number of 2n=38. Meiosis I and II stages were generally normal in the three orchid species although laggards and non-congression of chromosomes were observed at varying frequencies. The highest frequency of laggards was noted during metaphase II in D. secundum f. album and P. bicolor (9.96 % and 17.91 %, respectively) and during telophase I in V. lamellata var. boxalli. Non-congression of chromosomes was most frequent at telophase II of D. secundum f. album and P. bicolor (10.96 % and 11.84 %), respectively) and at anaphase II of V. lamellata var. boxalli (10.31 %). Pollen fertility was high in all three species due to the relatively high frequency of normal chromosome behavior - 85.47 % in D. secundum f. album, 81.46 % in P. bicolor, and 75.22 % in V. lamellata var. boxalli.
