Abstract
To differentiate Lacatan cultivars of banana (Musa sp.) from Cavite, Davao, and Mindanao, and Lacatan putative hybrid, isozyme analysis was performed through starch gel electrophoresis. Twelve enzyme systems, namely: acid phosphatase (ACP), alkaline phophatase (ALP), esterase (EST), glucose-6-phosphate dehydrogenase (G6PD), alpha-2Iycerophosphate dehydrogenase (GPD), glutamate oxaloacetate transaminase (GOT), isocitrate dehydrogenase (ICD), malate dehydrogenase (MDH), malic enzyme (ME), phosphogluconate dehydrogenase (PGD), glucose phosphate isomerase (PGI), and shikimate dehydrogenase (SHTK) were used to detect unique isozyme bands and band patterns. None of these enzymes recognized unique bands and banding patterns to differentiate the three Lacatan cultivars and the Lacatan BBTV putative resistant hybrid. Equally important, the Lacatan cultivars and hybrid displayed a common banding pattern with the two known cultivars seen in ICD, MDH, ME, PGI, and SHIK, indicating little or no biochemical variation among the Musa collections. Moreover, in ACP, ALP, GPD, GOT, and PGD enzyme systems the Lacatan cultivars and hybrid exhibited distinct banding pattern from that of Bungulan (AAA) and Latundan (AAB), eliminating the probability of having an AAA or AAB genome.
