Abstract
Evaluation of the methionine content of individual seeds of interspecific F2 hybrid populations of mungbean ( Vigna radiata (L.) Wilczek) and blackgram (Vigna mungo (L.) Hepper) was conducted at the Nutrition and Analytical Laboratory of the Asian Vegetable Research and Development Center (AVRDC). Analysis of the methionine content was performed using the modified methionine bioassay. Usually, the method would take one week to accomplish but by simply organizing the incubation periods of the culture used, it was reduced to three days. Other modifications in the method were the use of sterilized water for extraction instead of ethanol, the use of HC1 and NaOH, and heating to avoid contamination. Several experiments were performed first before measuring the methionine contents of the hybrids to determine the sample size to use. Results showed that the best sample size to use is 5mg. It was also seen in other experiments that there is a large variation of methionine content among the seeds of the same line. Such intraline variation was exhibited by the seeds of mungbean (9.6+1.0), VM 2164 (59.2+2.4), and G x B no. 2 (53.4+4.1). F2 interspecific hybrids of mungbean, VM 2164 (blackgram) and GxB no.2 namely VMC 01001-2B, VMC 01001-6B, and VMC 01002-6B were observed to contain more methionine content than their parents. This can be explained by the occurrence of transgressive segregation. To determine the methionine-rich seeds to select, the frequency distribution of the hybrids for their methionine content was graphed. The seeds with higher methionine than the parents were selected. Methionine bioassay still needs further modification to improve its accuracy. However, in the meantime, such problem was remedied by determining the methionine contents of the parents for each batch of screening.
