Abstract
GAYETA, CLAUDIO II QUIMO. April 2005. Genetic Polymorphism in Various Philippine Trichoderma Isolates Based on Random Amplified Polymorphic DNA. University of the Philippines Los Banos. B. S. Biology, major in Cell Biology. Major Adviser: Ma. Genaleen Q. Diaz, Ph. D. ABSTRACT Genetic polymorphism in various Philippine Trichoderma isolates were analyzed using Random Amplified Polymorphic DNA (RAPD) technique. Genomic DNA were isolated from 15 samples collected from different provinces of the Philippines. Five primers (AMO2, AM03, AMOS, AM07, and AM 10) were used to carry out the amplification process. A total of 70 polymorphic hands were obtained in all the isolates using the live RAPD primers. The highest computed value for gene frequency was 0.8000 for loci AM03-5 and AIMS-4, wherein 12 among the 15 samples (80%) exhibited the presence of bands. The computed average value of genetic variation was 0.2938 with a standard deviation of 0.1310. Isolates 'CDO' (T pseudokoningii) and '136' (T harzianum) gave the lowest genetic identity value of 0.5286 and the highest genetic distance value of 0.6376. Isolates 'HAPS' and 'Horturium', as well as '137' and 'Horturium' (both pairs are isolates of T harzianum) gave the highest genetic identity (0.8000) and the lowest genetic distance (0.2331). A dendrogram derived from genetic distance showed that Hingyon (7'. harzianum) is the most genetically distant among all the samples.
