The study was conducted to develop a technique to isolate cells from anthers of Saccharum spontaneum L. in large quantities and test for their viability. The technique developed yielded an average of 5,000 cells /ml (range was from 4,000 to 7,000 cells per ml). Higher density of cells could be obtained but they are of less purity. The best number of filter layers was six using gauze bandage as the filter material. The length of centrifugation time significantly affected the density and purity of isolated cells. Twenty minutes was the best where relatively many cells with high purity were isolated. Younger tissues were found to yield less isolated cells than older ones.
