ABSTRACT To clone and partially characterize the Acyl Carrier Protein (ACP) gene involved in fatty acid biosynthesis in coconut. cDNAs of the putative ACP gene isolated from a 4-month old (m.o.) coconut endosperm was amplified using 3' RACE (Rapid Amplification of cDNA Ends) and was cloned. The sequence obtained contained 488 bp, which was more than the expected size of 300 by as previously cited. Analysis revealed that only 56.6% of the primer annealed during amplification. Moreover, there was a 97 by region before the primer. BLASTX analysis of the region before the primer and the region from the primer to the poly-A tail showed alignment with genes coding for proteinases, endopeptidases, and vacuolar processing enzymes. Based on this, it cannot be concluded that the clone is the putative ACP gene. However, BLASTX results for the whole sequence including the insert had an alignment with the predicted Coding Strand (CDS) ACP of Caenorhanditis elegans. Out of the aligned 37 amino acid residues. 34% were identical amino acids and 57% similar amino acids. This suggested that the putative ACP might have been cloned in this study, but because of the action of the topoisomerase I, the sequence of the gene was altered, and consequently its homology with the ACP gene sequences of other organisms.
