Abstract

HEMEDEZ, RIZALINA RACQUEL A. University of the Philippines at Los Banos. April ,1982 A Preliminary Study on the In Vitro Culture of Human Squamous Cell Carcinoma 4,1 Coconut Water Media. Adviser: Jr. William G. Padolina Collagen-dissociated human squamous carcinoma cells cultured for seven days at 37°C in Coconut Water(CW), Coconut Water with Serum (CWS), (both obtained from approximately six months old drupes) and Eagle's Minimum Essential Medium (MEM) exhibited epithelial cell sheets and monolayers with no contact inhibition on days two and three together with some fibroblasts and lymphocytes as confirmed by Papanicolaou staining. Type, blue dye-exclusion tests and acridine orange tests shored that these cells exhibit a characteristic growth curve with the highest growth rate on the second day for CW, third day for CWS and second and third day for MEM. Results suggest that CWS could be used as a substitute for MEM. Results suggest that CWS could be used as a substitute for MEM as a culture medium for carcinoma cells. Comparison of the chemical composition of Coconut Water against Eagle's Minimum, Essential Medium and other cellular growth requirements in vitro were discussed.
