Sodium-dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was used to examine seed storage proteins from Vigna radiata (L.) Wilzeck cv. NCM 53 and Vigna mungo (L.) Hepper cv. ACC 25. At least five polymorphic polypeptides were observed between NCM 53 and ACC 25 and their mode of inheritance were established upon examination of 102 F2 progeny of the cross. Loci 1 (L1) and 2 (L2) consist of a 67.7 KD and a 65.9 ED polypeptide, respectively, in NCM 53 and both null forms in ACC 25. The Segregation ratios for locus 1 deviated significantly from expected Mendelian ratios for single dominant gene. Locus 2, however showed a good fit to a 3:1 ratio. Locus 3 (L3) is composed of a 60 KD protein in NCM 53 and a 58.8 KD polypeptide in ACC 25. L3 showed a 1:2:1 segregation ratio typical of a single co-dominant allele. In Locus 4, (L4) NCM 53 possesses the 39.5 RD type while ACC 25 exhibited the null form. A 3:1 segregation pattern was observed favoring the 39.5 ED type thus indicating a single dominant gene action. Locus 5, (L5) consists of a 21.5 KD polypeptide in NCM 53 and a 20.8 KD polypeptide in ACC 25. L5 segregation pattern of 1:2:1, typify a codominant interaction of alleles in a single gene locus. the possible identities of the polymorphic bands were inferred.
