Abstract
MADRIGAL, APRIL JOY. APRIL 2004. MORPHOLOGICAL, CYTOLOGICAL, AND ISOZYME ANALYSIS OF TISSUE CULTURE-DERIVED CORN (Zea mays. L.) FROM IMMATURE EMBRYOS. University of the Philippines, Los Banos Laguna. Adviser: Merlyn S. Mendioro Morphological, cytological and isozyme analyses of tissue culture derived (TC) corn (Zea mays. L.) from immature embryos were done to explain the somaclonal variation noted on TC plants. Differences were noted in plant height and number of shoots. The control P53 and PI 17 measured 143 and 131.2 cm, respectively. The TC plant height ranged from 54.8 to 72.4 cm. Days to tassel and silk formation, number of leaves, and height of ear were higher in the control plants. Early ear initiation was noted in TC. In P53 and PI 17 it takes 47 to 57 days but in the TC initiation is from 34-49 days. Multiple ear formation, longer ears, multiple tillers were observed only TC PI 17. In TC P53, more seeds were produced as compared to P53. Sporocytes of P53 and TC P53 were studied cytologically. P53 and TC P53 showed normal diakinesis with 10 IIs. Metaphase 1, anaphase I, and telophase l were all normal. Pollen fertility is 100%. Isozyme studies were done on V4 (control) and four TC plant. Three isozymes, acid phosphatase (ACPH), alkaline phosphatase (ALPH), and esterase (EST) were observed through starch gel electrophoresis. Only one locus for ACPH1 was observed. Two genotypes were noted, FF and FS. For ALPH, two locus were observed (ALPH 1 and ALPH2). For ALPH1, genotypes SS, FS, and FF were observed. EST was monomorphic. The control and TC plants showed the same genotype (SS). Except for EST, TC plants showed polymorphism in ACPH and ALPH.
