BS Biology major in Genetics Major Adviser Prof. Celia B dela Viña ABSTRACT The chromosome behavior in meiosis 1 and pollen fertility of two sets of parents using thermosensitive genic male sterile (TGMS) lines of rice and inbred varieties (PSB Rc) and their doubled haploid (DH) regenerants were studied. Generally, cells of the parentals and their DH regenerants exhibited normal stages of meiosis I. Diakinesis showed a prevalence of 1211s; therefore the diploid chromosome number was 211=24 in all the plants. TGMS lines and DI-I lines showed some cells with univalents (1111 + 11, 1011 + 41) in addition to bivalents. Laggards and non-congression were observed at metaphase 1 (1-3% and 2%, respectively) and at anaphase I (1% and 1%, respectively) but at low frequencies. A very minimal frequency of abnormality (1-2%) such as laggards and non-congression were also documented at metaphase 1 and anaphase 1 in the regenerants from Fi of TGMS6 x PSB Rc98. These were likewise observed at metaphase 1 and anaphase I (1-11%) of the regenerants from F1 of TGMS13 x PSB Rc30 Pollen fertility analysis showed that most of the pollen grains, except for the inbred varieties, can be categorized as partially fertile to fertile during the whole 12-day duration after booting stage. The critical sterility point (CSP) for TGMS6 and TGMS13 lines were found to be 27.43°C and 27.5°C, respectively. For the regenerants 1, 2 and 3 from the F1 of TGMS6 x PSB Rc98, the critical sterility points were found to be at 28.51°C, 28.56°C and 27 64°C, respectively. Likewise, for the regenerants 1, 2 and 3 from the Fi of TGMS13 x PSB Rc30, the CSP were observed at 28 48°C, 29.1T and 28.38°C, respectively.
