Abstract

Mamaril, Albert C. University of the Philippines at Los Banos, April, 1982. Electrophoretic Patterns of Soluble Endosperm Proteins from 8-9 Month Old Makapuno and Normal Coconuts.

Advisers: Dr. Dolores A. Ramirez and Mr. Cesar V. Mujer.

The protein hand patterns of normal and makapuno coconut endosperms (8-9 mo), resolved independently by polyacrylamide gel electrophoresis (PAGE), sodium dodecyl sulphate-polyacrylamide gel electrophoresis and IEF, respectively resolved 16, 21, and 16 protein bands in the normal endosperm in contrast to 17, 18, and 19 bands, respectively, in makapuno. The patterns obtained after SDS-PAGE showed four heavily stained major bands in makapuno: Py1, Py4 , Py5 and Py6 with molecular weights of 16, 24.5, 30.5 and 48 Respectively. Similar bands were detected in the normal except for Py5 which was replaced by two slightly stained minor bands. Further-more, two minor protein bands Py2 (Mr = 16K) which was present in the normal but absent in Makapuno and Py3 (Mr = 21.8K) which was present in makapuno but absent in the normal were noted. The patterns obtained after IEF were basically similar between the normal and makapuno endosperms except for the presence of three additional bands in the latter PZ1, PZ2, Pz5 with pKI's of 5, 9, 5, 92 and 6...38 respectively
