Abstract

The effects of different concentrations of lead acetate (PbAc) on the unialgal culture of the green alga, Chlorella sp., were studied for its growth and morphology under laboratory conditions. Growth rate (doublings per day) of the alga for seven days showed a significant inverse relationship (a=0.05) to different lead concentrations ranging from 10ppm(TI), 2ppm(T2), 4ppm(T3), 6ppm(T4), and 10ppm(T5). Growth followed for 13 days showed that the five treatments generally had their peak of growth after seven days with the following treatments exhibiting decreasing order of growth: T I> T3> T2> T5> T4. The maximum growth was obtained in T I after seven days with absorption peak of 0.2915 at 680nm and the least was in T4 at 0.1496. At logarithmic phase of growth of the alga, its chlorophyll content, fresh and dry weight, and cell density were determined in the control (T1), as compared to 4ppm PbAc (T3) and 10ppm PbAc (T5) in the basal inorganic medium. Chlorophyll concentrations showed a decreasing trend with increasing amount of PbAc as follows: 5.5996mg/ml, 5.4845mg/ml, and 4.30664ml for T1, T3, and T5, respectively. 1-lowever, there was no significant difference (a=0.05) in the chlorophyll content in these treatments. There was also no significant difference among the treatments in their cell diameters whose sizes varied from 2.1 l3pm, 2.088mm, and 2.063mm for T1, T3, and T5, respectively. The cell density was highest in '13 (6,556,500 cells/ml), followed by T1 (5,658,000 cells/m1) and the least was in T5 (2,252,000 cells/m1). The fresh weight was highest in T1 followed by T3 and then T5 with values of 0.02880111, 0.0230g/ml, and 0.0222g/ml, respectively. The dry weight also had an inverse relationship to the different treatments with the following results: 0.0055g/ml for TI, 0.000105g/m1 for 13, and 0.000065g/m1 for T5. There was a decrease in the size and/or fragmentation of the chloroplast in 6ppm and 1Oppm PbAc as compared to the control (T1).
