Sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) was used to determine the seed protein profile and differences among Luffa cylindrica M.J. Roem., L. acutangula Roxb., and their F, hybrid. There were a total of 18, 19, and 20 repeatable polypeptide bands observed among 60 samples of L. cylindrica M.J. Roem., L. acutangula Roxb., and their F, hybrid, respectively. Band numbers 13(Rf = 0.6517) and 16(Rf = 0.7433) were found to be unique to L. acutangula while band number 10(Rf = 0.5366) is particular only to L. cylindrica. There were no unique bands found in the F1 hybrid but it exhibited the bands unique to each of its parents, indicating that it is indeed a hybrid. Due to the presence of only one dissimilar band (band number 10) between L. cylindrica and the Fi hybrid, similarity index (0.9500) is high compared to L. cylindrica and F1 hybrid (0.9000) which have two dissimilar bands (band numbers 13 and 16). Similarity index between L. cylindrica and L. acutangula shows that there exists a high similarity (0.8500) between the two species. The very little protein variation observed indicates considerable genome homology of L. cylindrica and L. acutangula.
