Abstract
Three strains of the domesticated silkworm, Bombyx mart L , namely Shoukou x Ryuuhaku, Guanong (G) and Kinshu x Shouwa (KS) were characterized using morphological and isozyrne analyses. Differences on larval crescent and star spot markings were observed between strains. SR has a crescent light brown inside and dark brown outside and a dark brown star spot. G has a crescent dark brown outside and light brown inside and a light brown star spot. KS has a crescent dark brown inside and light brown outside and a dark brown star spot. Five out of eleven traits were significantly different among strains namely egg count per egg mass, hatchability, normal and abnormal moth emergence rates and number of eggs laid. Isozyme analysis using starch gel electrophoresis for acid phosphatase (ACPH), alkaline phosphatase (ALPH), esterase (EST) and malate dehydrogenase (MDH) revealed sixteen presumptive loci. All presumptive loci were polymorphic except ACPH-3, ACPH-4, ALPH-4, EST-1 and MDH-3. ACPH-3S, ACPH-4S, ALPH-1M, ALPH-4F, MDH-2S and MDH-3S were unique for SR while ACPH-1F, EST-1S and MDH-1F were unique for KS but none for G. Likewise, genotypes ACPH-2MM, ACPH-2FF, ACPH-3SS, ACPH-4SS, ALPH-1MM, ALPH-4FF, MDH-2SS AND MDH-3SS were unique for SR; genotypes ACPH-2FS, EST-4MM and EST-4FF for G and genotypes ACPH-1 FF, ACPH-2FM, ALPH-3FS, EST-1SS and MDH-1FS for KS. Similarities within and among strains as measured by the Jaccard's similarity (SI) showed that individuals within KS had a higher SI value of 47.334 than SR and G with SI values 30.722 and 30.58 respectively. A comparison between SR and G strains gave the highest SI value of 17.56 as compared with between SR and KS and between G and KS with SI values of 10.4 and 11.9, respectively.
