Abstract

Normal filaments consisting of an average of 39 cells per filament were observed in 0 age level cultures. This filament had a basal heterocyst. Akinetes were observed in all cultures incubated for 17 days in distilled water and phosphorous-deficient media subjected to darkness, white light and blue light. Akinete formation occurred adjacent to the heterocyst. A mature akinete was larger in size, had a thicker cell wall and granular cytoplasm. Seventeen day old cultures containing an akinete per filament with no vegetative cells had lower amounts of chlorophyll a and protein compared to 0 age level cultures. On the other hand. dry weights were higher in 17 day old cultures. After two weeks of incubation in inorganic medium depleted of nitrogen in vitro, akinete started to differentiate into vegetative cells. The germinating akinete elongated further into a normal filament. Chlorophyll a, protein amounts and dry weights increased after germination. The process of germination seemed to be affected by the presence of nutrients such as phosphorus and light.
