Electrophoretic patterns of soluble endosperm proteins from 10-11 month old normal makapuno coconuts

Abstract

Proteins extracted at pH9 from 10-11 month old normal and makapuno endosperms were subjected to electrophoresis (PAGE). sodium dodecyl sulfate electrophoresis (SDS-PAGE) and isoelectric focusing (IEF). Protein band patterns showed that one protein in PAGE, four in SDS-TAGE and eight in IEF were unique to the normal endosperm. Six proteins in PAGE. five in SDS-PAGE and nine in IEF were exclusive to the makapuno endo-sperm. One protein in SDS-PAGE, two in IEF and none in PAGE occurred in both endosperms but in higher intensity in the normal. Two proteins in PAGE. three In SDS-PAGE and six in IEF had higher intensities in the makapuno endosperm. PI values of bands in PAGE molecular weights of bands in SDS-PAGE and isoelectric points of bands in IEF were computed. The differential protein incidence in the two endosperms suggests a genetic misregulation of biosynthetic pathways for proteins in the mutant endosperm.
