Isozyme analysis in stingless bees, Trigona biroi Friese, from Laguna, Albay and Cavite using starch-gel electrophoresis

Abstract
Genetic variability in stingless bees, Trigona biroi Friese from Los Banos, Laguna; Guinobatan, Albay; and Silang, Cavite was analyzed using starch-gel electrophoresis. Among 180 samples analyzed, six presumptive loci were observed encoding for four enzymes: two loci each for acid phosphatase (ACPH) and alkaline phosphatase (ALPH); one locus each for esterase (EST) and glucose-6-phosphate dehydrogenase (G-6-PD). Two codominant alleles: S (slow) and F (fast) were coding for ACPH-1, ALPH-1 and EST while only the F allele controlled ACPH-2, ALPH-2 and G-6-PD. Enzyme polymorphism was observed for ACPH-1 and EST in all populations while ALPH-1 was polymorphic only in Laguna. ALPH-2 was monomorphic and was found only in Cavite while ACPH-2 and G-6-PD was monomorphic in all populations. The proportion of polymorphic loci, average number of alleles per locus and average heterozygosity were highest in Laguna, followed by Cavite and lowest in Albay. These indicated that Albay population was least diverse while Laguna population was most diverse. Laguna and Albay showed the highest degree of genetic identity (0.977) and genotypic similarity (0.922). Lowest value for genetic identity (0.772) was obtained between Laguna and Cavite while genotypic similarity was lowest (0.594) between Albay and Cavite.
