Abstract
Protoplasts were isolated from the spindle tissue of Saccharum officinarum using an enzyme mixture of cellulase, sorbitol and magnesium chloride. The effects of cellulase concentration, pH and sorbitol concentration were investigated. Cells at different stapes of cell wall degradeticn were observed. Protoplasts isolated were irregularly shaped due to the high sugar concentration used as storing medium. Optimum results were obtained at 4% cellulase, pH 5.0 and 0.5 M sorbitol.
