Abstract
The genetic diversity of Philippine Aglaonema species was determined based on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of leaf proteins. Isozyme analysis using alkaline phosphatase, acid phosphatase, and esterase enzyme systems were also done to further establish the genetic polymorphism of Aglaonema Using SDS-PAGE, a total number of 28 hands were resolved. Distinct banding pattern was observed among the cultivars of Aglaonema commutatum var. commutatum. Band similarities were also observed among the cultivars of A. commutatum var. picturatum. Among varieties, similarity is low with highest similarity index (SI) value of only 0.45. Comparing the different species, the SI values obtained ranged from 0 to 0.21 which implies that genetic diversity between species is high. Common banding patterns were also observed among the unclassified Aglounema cultivars. Between the twenty cultivars, the SI values were generally low ranging from (1 to 0.08. However, many of the cultivars have also high similarity index value ranging from 0.5 to 1.0. Among the cultivars with similar banding patterns identified which has an SI of 1.0 were White Rajah and Malay Beauty; Manila Whirl, Tricolor and Manila. Considering the three enzymes, results of starch gel elctrophoresis revealed polymorphism in the studied population. Alkaline phosphatase produced two alleles in its locus, while acid phosphatase has two loci and a total of five alleles. There were three loci identified in the esterase enzyme system and a total of 7 alleles. It was also observed in the three enzyme systems that cultivars under A. commutatum var. commutatum show consistent band similarity. Among the unclassified Aglounema cultivars, Manila Whirl, Manila and Tricolor ()lien show similar banding patterns in the three enzyme systems.
