Optimization of conditions for the immobilization of invertase of fine white lahar: 3- aminopropyltriethoxysilane, glutaraldehyde and invertase concentrations

Abstract

The use of white fine lahar as an organic support in the immobilization of invertase was investigated. Invertase immobilization and its broad applications in continuous processes are attractive, allowing the production of a final product with high purity which is typical of an enzymatic process, and without liberation of toxic products. the porosity of lahar makes it applicable as an organic support. Lahar samples were obtained from Botolan, Zambales. Upon optimization of APTS and glutaraldehyde concentrations, the values obtained are 10% and 3%, respectively. The lahar was silanized using the optimum APTS and activated by the optimum glutaraldehyde concentration obtained. Attachments of APTS and glutaraldehyde were confirmed using chemical tests and analysis. Attachment of APTS was confirmed using Hinsberg and nitrous acid tests while the attachment of glutaraldehyde was further confirmed using 2, 4-DNP and Tollens tests. A 0.2 mg.ml. invertase solution was immobilized to the white lahar using 10% AP'I's and 3% glutaraldehyde. The immobilized invertase exhibits maximum activity at pH 4.5 at 45 °C using 0.3 M sucrose solution while the free invertase gave maximum activity at pl 1 5.0 at 55° C using 0.3 M sucrose solution. The kinetic parameters were obtained for both frcc and immobilized invertase. The V„,,„ for free and immobilized invertase was 14.32 (pmol/min) and 10.08 (prnol/min), respectively. The Kr„ of the free invertase (0.023 mM) was lower than the Km of the immobilized invertase (0.369 mM). The one-week old immobilized invertase shown decreased in activity and remains its stability at pH 4.5 and at 45°C.
