Abstract
A crude enzyme mixture from the haustorium of germinating coconut (Coco nino variety) was extracted using 0.05M phosphate buffer pH 7.5. The extract has a protein content of 8.6 mg/mL determined by the Lowry method. Using coconut oil as substrate, the crude enzyme extract had optimum lipolytic activity at pH 7.0, 45 deg C, and 20 minutes reaction time. Ninety percent of the lipolytic activity remained until the sixth day of storage at 10 deg C. Activity dropped sharply on the eighth day and was no longer detectable on the 17th day. Gas chromatographic analysis revealed that caprylic acid is preferentially liberated from the hydrolysis of coconut oil by the action of crude enzyme extract from the haustorium of germinating coconut, comprising about 95% of the total free fatty acids hydrolyzed.
