Analysis of the ascorbic acid content of dalandan citrus fruit, Citrus nobilis Lour, using HPLC

Abstract

The ascorbic acid content of the dalandan fruits obtained from Batangas and Davao were determined by making use of Iligh Performance Liquid Chromatography (11PLC). Three methods of extraction were employed: (1) normal manual extraction. (2) manual extraction using a squeezer. and (3) automatic extraction using a blender. their effectiveness were examined and evaluated through analysis using HPLC

External calibration was done for the quantitation of the ascorbic acid content of the dalandan fruit samples by HPLC. For both the standards and the samples. the peak area and the peak height measurements were obtained and compared. Although their values were quite close, the peak area salues were more precise than the peak height values. Results showed that using the automatic extraction would give a higher ascorbic acid level compared to the two manual extractions. For the dalandan fruits acquired from Batangas and Davao, values of 1 1 l ± 3 and 143 ± 2ppm, respectively, were obtained from the automatic extraction while for the manual extraction and mechanical extraction using a squeezer, values of 94.2 ± 1.5 and 139 ± 3ppm, 95.9 . 2.0 and 145 ± 2ppm were obtained respectively.
