Abstract
This study aims to develop a laboratory scale preparation Procedure for the extraction and purifica-tion of malate dehydrogenase from beef heart using dye-ligand chromatography system and to screen for a two-column, negative-positive dye-ligand system for its purification. The 60-200 mesh rehydrated particle size nata de coco was used as the solid support and different dyes, namely, Procion Red HE-3B, Procion Blue K-EG, Procion Yellow MX-GR, Procion Turquoise H-A and Procion Green HE-4BD were used as dye ligands. Procion Red HE-3B-nata was chosen as the negative column with 69% recovery and 2.2 fold pure. The pooled fraction from the negative column was passed through the positive column which is Procion Green HE-4BD-nata. Elution by step gradient concentration of NaCl was carried out and gave 56% recovery and 60.6 fold pure from the two-step purification process
