Abstract
The use of lahar as an inorganic solid support for immobilization of yeast invertase was evaluated. Yeast invertase was covalently immobilized on activated "lahar". A -lahar" sample was fast ignited at 550 °C for 5 hours followed by treatment with concentrated hydrochloric acid, where the two components of lahar (white and black) were separated_ The two components were activated by treating with y-aminopropyltrietho;cysilane followed by reaction with glutaraldehyde. The activated labor was then incubated with invertase. The covalent coupling of silane and glutaraldehyde onto lahar were verified by chemical tests and FT-IR analyses. Invertase covalently coupled with lahar retained about 76.5% of its activity. Storage stability of both immobilized and free enzyme were compared and results show that the stability of the former was enhanced The activity of the immobilized enzyme was relatively stable towards repeated use.
