Extraction characterization and comparison of piscicidal activities of rotenoids from Derris elliptica and Derris trifoliata

Abstract
Rotenoids from Derris elliptica and Derris trifoliata were obtained by extraction with acetone. To further characterize, the acetone extracts were partitioned in chloroform and water where the chloroform extracts yielded a dark brown residue. Chromatographic profiles of the said extracts were obtained through High-Performance Liquid Chromatography (HPLC) using a Ci8 bonded reverse phase column and 60:40 (v/v) methanol:water as mobile phase. Sample extracts were dissolved in acetonitrile prior to injection, and HPLC fractions were collected. Liquid Chromatography-Mass Spectroscopy (LC-MS) was employed to obtain the mass spectra of these LC fractions. From these mass spectra, the components of the extract were classified into two rotenoid groups; the group containing the m/e 392 or 394 and the group containing m/e 410. Six HPLC fractions were collected from the chloroform extract of D. elliptica and five HPLC fractions were collected from D. trifoliata. Fractions El, E2, Ti, T2, T3 and T4 belong to Group 392-394 and fractions E3, E4 and E6 belong to Group 410. Group 394 fractions were found to be deguelin or rotenone and 392 fractions were found to be either dehydrodeguelin or dehydrorotenone while Group 410 fractions are found to be one of the following: sumatrol, tephrosin or toxicarol. Dehydrorotenone were identified definitely in fractions El and T2 and dehydrodeguelin were identified in fractions E2 and T3. Also, rotenone was identified in T1. Finally, the piscicidal activities of the chloroform extract, acetone extract, aqueous extract and crude extract from both species were determined using tilapia (Oreochrornis nilotica) fingerlings for the bioassay. Results. demonstrated the relative toxicity of the said extracts and their potential use as piscicides.
