Optimization of conditions for acid protease production by solid substrate fermentation using Rhizopus oligoporus BIOTECH 3094 was conducted. The variables analyzed in this study were: substrate = rice bran, corn cobs and wheat bran, incubation time = 0 to 11 days, incubation temperature (°C) = 27- 30 (RT), 37, 45 and 50, and initial pH of the mdium = 2.0, 2.5, 3.0, 3.5, 4.0 and 4.5 The optimized conditions for acid protease production were as follows: substrate = rice bran; initial pH of the medium = 2.5 ; inoculum level = 5 % (v/v), incubation temperature = 27 - 30 ° C (RT) and incubation time = 4 days.

Comparison of the unoptimized and optimized conditions for bench scale production of acid protease using the above organism was conducted. Data showed that the specific activity (U/mg protein) was higher using the optimized conditions than the unoptimized conditions, 0.565 (U/mg protein) and 0.513 (U/mg protein) respectively.
