Bacillus subtils BIOTECH 1332 and Bacillus amylohquefaciens BIOTECH 1583 were selected for bench scale xylanase production under neutral and acidic pH, respectively, for their ability to exhibit high xylanase activities.
The optimum incubation periods determined for bench scale xylanase production of 11..cruhtrlis BIOTECH 1332 and B. amyloliquefaciens BIOTECH 1583 were 24 and 36 h, respectively. For the incubation temperature, the optimum xylanase activities for both bacterial isolates were obtained at room temperature. Both bacteria gave highest specific xylanase activities at 200 rpm agitation rate. The optimum aeration rate obtained for B. =kills BIOTECH 1332 was at 0.5 vvm while for B. amyloliquejamens BIOTECH 1583 was at 1.5 vvm. Bacillus subtilis BIOTECH 1332 gave highest xylanase specific activity at pH 7 while B. amyloliqucfaciens BIOTECH 1583 had highest activity at pH 5.
The optimized conditions resulted in significantly higher xylanase specific activities than the unoptimized conditions for both B. subtilis BIOTECH 1332 and B. amyloliquefaciens BIOTECH 1583 at 227.87 and 144.74 XAU/mg protein, respectively.
