Bioassay of the makapuno milk: cytology of carrot tissue cultures

Abstract
The macapuno milk was unable to stimulate in the culture carrot tissues a tumorous condition similar to that of the macapuno endosperm. Although haphazard cell arrangement and orientation was found in the car- rot tissues, microcells as well as crisscrossing cells and cell wall and the lack of intercellular adhesion were not observed.
It was further observed that in the 4-week culture of the carrot tissues, the milk of normal coconut from macapuno bearing tree had similar effects to that of the macapuno milk. On the other hand, the effect of the normal coconut from normal tree was similar to that of the milk of immature coconut. However, the effects of the milk of macapuno and normal coconut from macapuno- bearing tree were different from that of the milk of nor- mal coconut from normal tree and milk of immature coconut. The tissues in the cultures supplemented with milk from macapuno and normal coconut from macapuno bearing trees have bigger cells (length and area) which are more elongated compared to those in medium supplemented with milk of normal coconut from normal tree and milk of immature coconut. In the eight-week culture the different treatments had similar effects indicating a levelling off of the active fraction of the supplements resulting in similar effects on the cells of the carrot tissue culture.
