Abstract

B-Casein is a major lactoprotein with two most common variants in cattle, these variants only differ in the 67th protein at the 7th exon. These variants are A1 and A2 where A1 is suspected to be linked with diabetes, coronary heart disease, arteriosclerosis and sudden infant death syndrome. Frozen-thawed semen were used as samples for the PCR-based screening of β-Casein. The whole semen collection, processing and preservation were done at the Philippine Carabao Center at UPLB. DNA from frozen-thawed semen were extracted and amplified. The amplified products were then sent for sequencing however the sequences of the DNA were erroneous which might be due to the PCR products not being concentrated enough, the DNA templates used to have a very low concentration and the frozen-thawed sperm cells experienced double standard DNA breaks due to cold injury which affected the sequencing process which is fluorescent based.
