Abstract

The research internship was conducted at Food Science Cluster Department under the College of Agriculture of the University of the Philippines Los Baños, Laguna from April 21, 2014 to December 4, 2014. The internship was about to test the prebiotic activity of QPM corn grits. This experiment exposed the student to different techniques that can be applied in the actual working condition in the laboratory. Learned techniques helped the researcher attained the results of this study. The experiment to test the prebiotic potential of QPM corn grits includes the use of prebiotic assay and prebiotic activity score. The ground corn grits in 0.5%, 1.0%, 1.5% and 2.0% (w/v) were added in MRS broth to test its prebiotic activity with Lactobacillus acidophilus and Lactobacillus casei. Escherechia coli were grown in M9 minimal medium containing concentrations of corn grits employed in Lactobacilli. Glucose served as the control while Yacon and FOS served as the positive controls. Cell densities of microorganisms in 0-hour were compared to the cell densities after 24-hour incubation. Cell densities were used to compute for prebiotic activity scores of different carbohydrate source. pH and %TA were also determined. Based on the obtained data, corn grits did not show a prebiotic potential. Through this internship, the researcher gained the competence to be part of food biotechnology industry in the future.
