Abstract
Twenty-five soil bacterial isolates were screened for IAA production using colorimetric assay. Five isolates (MCM11, MCM13, MCM15, MCM18, and MCM20) had high IAA production. Cytokinin was extracted from these isolates and with two others from MykoPlus Laboratory, SF5 and JLBB19. Cytokinin activity in decreasing concentration of extract was examined using leaf disk bioassay. After five days, JLBB19, MCM18 and MCM20 consistently had high cytokinin activity based on leaf extract absorbance. The isolates were characterized morphologically. The 16S rDNA was amplified though it was not successful. To further increase IAA production, the isolates were mutated using HCl and colchicine. After mutation, MCM13, MCM15, and MCM20 had significantly increased their IAA production. The strains were separately inoculated on corn and eggplant to check the effect of cytokinin- and IAA-like substances in growth. Shoot length, root length, chlorophyll concentration, dry weight and germination rate were noted after three weeks. MCM18 had the highest cytokinin-like activity among non-mutants for both host plants. Among acid-mutants, MCM13 on corn and MCM15 on eggplant had the highest cytokinin-like activity. Among the colchicine-mutants, MCM13 had the highest cytokinin-like activity for both host plants.
