Abstract
A PC-based automation module was constructed and installed on a manual compound microscope to quantify the infection of rice sheath blights caused by Rhizoctonia solani Kühn using digital image processing. The focusing and positioning knobs of the compound microscope were adjusted using calibrated stepper motors controlled by a microcontroller. The light source of the microscope is an array of LEDs that is also controlled by the microcontroller. The image captured by the camera is the
Rhizoctonia solani Kühn on the rice tissue specimen. It was quantified using a color- based segmentation algorithm. Also, a Graphical User Interface (GUI) was designed for
controlling the compound microscope and for applying the image processing algorithm for quantifying Rhizoctonia solani Kühn. The performance of the automated microscope was tested by comparing its measurement of the infestation with the measurement made by humans using manually operated microscope. The latter tended to overestimate the amount of infestation. This is due to the effect of gestalt principle of human perception. This could be further minimized by improvement on the image focusing and specimen preparation.
