Abstract
Extraction of oil from Chlorella vulgaris harvested from 10000-L raceway pond using methanol-chloroform (1:2 v/v) was investigated at different biomass-to-solvent ratio (1:6, 1:10, 1:14 g:mL) and soaking time (8, 16, 24 hours). The results were analyzed using Two-Way ANOVA with replication and was found out that only the biomass-to-solvent ratio has significant effect on the oil yield: when the solvent is increased, the oil yield also increased.The highest oil yield result is 15.8763% at 1:14 ratio and 24 hours soaking time. The oil was characterized using Thin-Layer Chromatography and BIOSOFT Quantisanᵀᴹ program. The effects of the parameters were also investigated on the free fatty acid (FFA) and triacylgleceride (TAG) contents of the oil. The parameters have no significant effect on the TAG content while biomass-to-solvent ratio has a significant effect on the FFA content: as the solvent is increased, the FFA content decreased. The overall average of the components are 16.86% TAG, 58.71%FFA, 10.34% 1,2-diacylglycerides (DAG), 5.22% 1,3-DAG, and 8.87% monoacylglycerides. Soxhlet extraction was also done at 16 hour operation with 1:14ratio which yielded an average oil yield of 15.25% with average oil contents of 23.55% TAG, 67.19FFA, 2.47% 1,2-DAG, 1,3-DAG, and 3.16% MAG.
