Abstract

The occurrence, identity, free-radical scavenging and anti-inflammatory activities of phenolic compounds in lima beans (Phaseolus lunatus L.) were determined. A descending two-dimensional paper chromatographic technique was used to determine the phenolic profile of the phenolic extract in lima beans. The phenolic present were characterized according to their color under visible light, visible light with ammonia vapor, UV (366nm) light, UV (366nm) light with ammonia vapor, chromatographic mobilities, UV and IR spectra. The major phenolics present were quercetin, cinnamic acids and a cinnamic acid ester. The total phenolic content of the extract was determined using the Folin-Ciocalteau reagent. The phenolic extract was found to contain 22.1 ± 1.59 mg GAE/100g sample and 27.0 ± 2.24 mg QE/100g sample. The free radical scavenging activity of the phenolic extract at various concentrations was determined using the DPPH radical. The IC50 for lima beans extract was 490.03 ± 27.57 µg/ml. The anti-inflammatory activity of lima beans was determined using an in vivo CAM irritation assay using 10-day old duck eggs. At 150 µg dosage, the anti-inflammatory activity of lima beans was comparable to that of the NovaSoy soybean supplement (SS). At the same dosage, hydrocortisone and quercetin exerted a significantly higher anti-inflammatory activity than SS and lima bean extracts. In conclusion, lima bean extract contains a significant amount of phenolics. Lima bean extract has a good free radical scavenging activity that is better than the NovaSoy soybean supplement. It also has an anti-inflammatory activity that is comparable to the NovaSoy soybean supplement. Lima beans, an indigenous legume in the Philippines can be a good alternative to other legumes especially soybeans which are consumed for their health benefits.
