Abstract
Primary cultures of chicken embryo fibroblasts (CEF) derived from Philippine native Banaba chicken (BNB) and Rhode Island Red (RIR) were successfully established by using the warm trypsin culturemethod. Cell morphology, viability, confluence and dynamic growth were observed and differences between growth of cells in media with 5% and 10% fetal bovine serum (FBS) were analyzed. The results showed that the cells were healthy showing characteristic fusiform and fibrous structure and reached 95% confluence at 72 hours. Cell viability ranged from 84-98%. All growth curves appeared as typical ―S‖ shape curves which indicate that cells underwent the lag, log and stationary phase. The population doubling time was determined to range from 52-63 hours. Four passages of primary cells (5% FBS) were successfully conducted wherein the second passages were cryopreserved. Independent t-test (P<0.05) showed that there were no significant differences in the mean cell concentration and viabilities comparing RIR and BNB treatments as well as cells in media with 5% FBS and 10% FBS. This study has shown the morphological and growth characteristics of CEF from Philippine native Banaba chicken. The methods described in this study will be useful for future virus and genetics research.
