Abstract

Laminarin and mannitol are two carbohydrates extracted from Sargassum spp. Laminarin was further hydrolysed using Trichoderma longibrachiatum. To optimize the fermentation process, parameters such as substrate concentration ratio, incubation temperature and pH of the medium were varied. Best substrate ratio and fermentation parameters were identified through Response Surface Methodology: Central Composite Design. Design-Expert 7.0.0 (Trial Version) calculated the optimum incubation temperature and pH to be 35°C and 4.45, respectively. Substrate concentration ratio was found tobe insignificant. A confirmatory run was also conducted using these optimum parameters to test the validity of the Response Surface Equation, and a percent difference od -0.972% was obtained, thus the equation is valid. Lastly, using One-way ANOVA it was proven that there is no significant difference in using commercial mannitol instead of crude mannitol in ratio with laminarin for bioethanol production.
