Abstract
The DNA extraction procedure developed by Hoisington et al., (1994) has been successfully adapted for locally developed inbred lines of maize. With minor modifications made to suit local laboratory conditions, the yield and quality of DNA isolated using the aforementioned procedure was found adequate for RAPD (random amplified polymorphic DNA analysis).
Similarly, a previously protocol for RAPD analysis (Heun and Helentjaris, 1993) was successfully tested after monir modifications of magnesium chloride and template DNA concentrations. However, applicability of the RAPD conditions established in this study is limited only one primer and one DNA sample. More painstaking work needs to be done to establish the optimum RAPD conditions suited to local conditions. Problems of reproducibility and wide applicability needs to be addressed before RAPD technology can be successfully integrated into existing corn breeding program.
