Growth kinetics of rhizopus chinensis saito BIOTECH 3273 in the production of microbial rennet using coconut paring cake as substrate

Abstract

The growth kinetics of Rhizopus chinensis BIOTECH 3273 using coconut paring cake for the production of microbial rennet was evaluated. Flasks containing 4%, 6%, 8% and 10% by wt. concentrations of coconut paring cake (CPC) were placed in a rotary shaker set at 33 °C and 100 rpm for 3 d. The amount of microbial rennet produced for each concentration was determined by noting down the milk clotting time. Milk clotting times for all CPC concentrations were not significantly different from one another. Milk clotting time was also lowest on the 2nd day of fermentation. For the growth kinetics study, 4%, 6% and 8% CPC concentrations were placed in a fermenter set at 30 °C and 250 rpm. Samples were taken every 3 h for 48 h and were analyzed for biomass, initial total sugar concentration and milk clotting time. Lower clotting times were obtained at higher CPC concentrations. Of the growth models tested, the Tessier model was found to best fit the growth of R. chinensis. Growth of the organism was not inhibited when CPC concentrations were increased. Values of the kinetic parameters were 15.674 h-1 for maximum growth rate (μmax) and 150 mg mL-1 for affinity of the microorganism to the substrate used (KS).
