WA-CMS fertility restorer gene restores fertility of a cytoplasmic male sterile line of rice (Oryza sativa L.) developed from a mutagenized source

Abstract
The inheritance and the location of the Rf gene governing the pollen fertility restoration of IR73328A, a cytoplasmic male sterile (CMS) line with a mutagenized cytoplasm source, were determined. The BC 1 population from IR73328A *2/IR73330-83-1-2R cross composed of 202 individuals was evaluated for pollen and spikelet fertility. Bulked segregant analysis was performed to screen for polymorphic simple sequence repeat (SSR) markers to be utilized in the construction of the molecular map of the Rf genes. The data from polymorphic markers along with the pollen fertility data were used to construct a linkage map with Mapmaker. Interval mapping performed with Mapmaker/QTL revealed that a fertility restorer gene is on the long arm of chromosome 10. It is flanked by SSR markers, RM6132 and RM171, in chromosome 10 at a distance of 3.0 cM and 1.5 cM, respectively (logarithm of odds, LOD = 13.08). Another marker RM1108 was also mapped to be 4.6 cM from the gene. This is the same marker mapped by Sattari (2004) to be co-segregating with Rf-4 which restores fertility in wild abortive (WA) and Dissi CMS lines. Further screening with the sequence tagged site (STS) marker S10019/BstUI used by Sattari et al. (2007) confirmed that the restorer line in this study possesses the characteristic restorer band expected from the Rf-4 gene. It indicates that Rf-4 which was a known restorer of WA-CMS can also restore a mutagenized CMS. It further implies that WA lines may be converted to alloplasmic lines based on mutagenized-CMS without the need to breed for new restorers.
