Purification and characterization of novel bacteriocins from lactic acid bacteria isolated from Philippine fermented rice-shrimp or rice-fish mixtures
Abstract
Various proteinaceous fermented foods native to Central and Southern Luzon, Philippines were screened for bacteriocin-producing bacterial isolates. Seventy-one out of several hundreds of colony-forming units isolated by agar plate streaking were found antagonistic to the indicator microorganism, Lactobacillus plantarum ATCC 14917, through direct assay. By "spot-on-lawn" assay of pH-neutralized culture supernatant, 9 isolates were confirmed to be bacteriocin producers. The bacteriocins of 2 isolates that gave the highest inhibitory activities, identified as Lactobacillus plantarum BS and Pediococcus sp. AA-5a, were purified by pH-adsorption/desorption method or gel filtration chromatography and reverse phase high performance liquid chromatography. The bacteriocins were found proteinaceous without carbohydrate moiety and had pH and temperature tolerance even at pH 2 and 9 and at 121 C for 15 min. They were also confirmed to be similar to pediocin by polymerase chain reaction. The spectrum of antimicrobial activity is narrow, i.e., only against type culture strains of L. plantarum, Pediococcus acidilactici, Leuconostoc mesenteroides and Listeria monocytogenes. The growth of L. monocytogenes during milk fermentation with Lactobacillus casei was inhibited by both bacteriocins unlike that of the control strain P. acidilactici S3.
