Abstract
A polyclonal antiserum against TMV was produced using SDS-PAGE purified TMV protein as antigen. The antibody response by the rabbit was monitored using indirect enzyme-linked immunosorbent assay (ELISA) and membrane immunobinding assay (MIBA). Sample sera were collected before every injection and diluted to 1:10, 1:100, 1:1000 and 1:10000 for MIBA. ELISA was able to quantitatively determine the production of antiserum in the rabbit through absorbance readings while MIBA gave only qualitative data on the progression of antibody response. ELISA results indicate a fluctuating trend in antibody production but an overall increasing response was obtained. The sensitive and specific antiserum produced was found to be still capable of detecting TMV in infected samples diluted at 1:1000 to 1:10000.
