Note: Establishment of cell suspension culture and induction of somatic embryogenesis in abaca (Musa textilis nee) tissue culture
Abstract
Proliferation of meristematic globules was observed at the basal region of regenerated shoots and enlarged buds/scalps suspended in the initiation medium consisting of modified Murashige and Skoog +1.0 ppm 2,4-D and 0.2 ppm zeatin. Cell suspension cultures were obtained from the meristematic globules which where maintained for several weeks in the initiation medium. There were instances when somatic embryos developed directly from the leaf sheaths, buds or scalps, whereas spontaneous secondary globular embryo formation was observed originating from meristematic globules. Browning of tissues was reduced with the addition of 20 ppm L-cysteine in the culture medium and frequent transfer of cultures during the first 2-3 weeks in the initiation medium.
