Discovery of a natural insecticide from Streptomyces sp. Strain PCS3-D2

Abstract
A natural insecticide was isolated and purified from crude extract prepared by organic solvent extraction from fermentation broth of Streptomyces sp. Strain PCS3-D2. Thin layer chromatography revealed that the crude extract contained a mixture of at least six compounds of different degrees of polarity. The non-polar component (P1-M) of the crude extract was purified by reverse phase high pressure liquid chromatography. The purified compound has a retention time of 9.178 min and a Λmax (lambda sub max) = 272 nm. Bioassay of P1-M revealed that it is highly toxic to third instar larvae of Asian corn borer (ACB) Ostrinia furnacalis. The median lethal dose (LD50) of P1-M to ACB is 0.011 ug/larva with fiducial limits ranging from 0.0035 ug/larva to 0.018 ug/larva. P1-M has a molecular weight of 494.30 g/mol. The chemical structure of P1-M was determined by NMR spectroscopy. P1-M has no compound of similar structure in the ChemSpider database.
