Optimization of DNA extraction protocol for Hoya mindorensis Schlechter

Abstract
Hoyas are popular ornamental plants originating from Southeast Asia. No publication on DNA extraction protocol on any Hoya genus is available. Several protocols were tested for suitability in DNA extraction of Hoya mindorensis Schlechter. A modified Angeles et al. (2005) protocol has been developed and proved to be successful in obtaining DNA products. This protocol utilized high-salt extraction buffer (2.0 M NaCl). The amount of b- mercaptoethanol concentration was modified to 0.5 M and the use of SDS was omitted. The DNA obtained through this protocol was amplified successfully through PCR and was able to produce distinct polymorphic SSR bands.
