Genetic structure of CSI population in the outbreak and non-outbreak areas using cytochrome oxidase 1 (CO1) gene

Abstract

The genetic structure of CSI populations collected from 10 provinces in the Philippines was investigated using mitochondrial cytochrome C. Oxidase 1 (CO1) gene sequences. Samples of coconut scale insect from Indonesia are used as reference for Aspidiotus rigidus Reyne. Multiple sequence alignment of the sequences generated a consensus of 454 bases. Three Aspidiotus species, namely Aspidiotus destructor Signoret, A. rigidus, and Aspidiotus excisus Green were identified through Basic Local Alignment Search Tool (BLAST) with verification using the 285 gene sequences in addition to the suggested 2% level species boundary of DNA barcode. Six, three and two haplotypes were identified as A. destructor, A. rigidus, and A. excisus species, respectively. Translated nucleotide sequences generated 151 amino acid sequences with distinct amino acids substitution among species. BLASTn and BLASTp results confirmed that the amplified fragment is part of the CO-1 gene region. Another species, Hemiberlisiapalma Cockerell which is also within the Tribe Aspidiotini, was also analyzed. Phytogenetic relationships inferred distinct grouping within tribe Aspidiotini including Aspidiotus species from outbreak and non-outbreak areas.
