Murihexocin C from the Leaves of Annona squamosa Linn. Induces Apoptosis in Human Colon Carcinoma Col 2 Cell Line

Abstract
The acetogenin murihexocin C was isolated from the leaves of Annona squamosa Linn. The structure was assigned based on 1H and 13C NMR and HR-ESI-TOF-MS. This is the first report of the compound from this plant. Murihexocin C exhibited significant cytotoxicity to human colon carcinoma Col 2 cells (IC 50 0.50 μg mL -1 or 0.79 μM) and normal Chinese hamster ovarian cells AA8 cells (IC 50 0.78 μg mL -1 or 1.24 μM). Murihexocin C (50 μg mL -1) induced extensive apoptotic cell death in Col 2 cells at 12 h as demonstrated by the TUNEL assay. Activation of the apoptotic effector caspase 3 was observed and a Bcl-2-like protein was noted to be down regulated.
