In vitro assessment and identification of fungi associated with vesicular arbuscular mycorrhizal fungi isolated from the rhizospheres of agricultural crops

Abstract

Enhancement and development of effective plant growth promoting inoculants demand the presence of a different set of attributes which can help in colonizing the rhizosphere. Fungi from the roots and soil rhizospheres of different crops with VAM inoculants are isolated and screened. Isolates that showed a positive result in phosphate solubilization were tested in vitro on IAA production assay. The total phosphate content available in the culture filtrate was measured by UV-vis spectrophometer at 420 nm following the vandate molybdate method while for IAA production assay, colorimetric determination using Salkwosky's reagent. The result showed that Aspergillus sp. (112) Sterilia mycelia (113) and Penicillium sp. (C5) showed the highest solubilization of Tricalcium Phosphate (TCP) in Pikovskaya's liquid medium, however, no isolates positively response in IAA production assay. Identification of isolates was done in the basis of cultural and morphological characterization. Fungal microorganisms which showed good phosphate solubilization are considered as the best candidates for the development of biofertilizer.
